Identity
Nonapeptide-1, marketed as Melanostatine-5, is a synthetic nine-amino-acid peptide used as a cosmetic brightening active under the INCI name Nonapeptide-1. What distinguishes it from the tyrosinase-inhibitor brighteners is where it acts: upstream, at the signalling step that switches pigment production on, rather than at the enzyme that carries it out.
Mechanism (as proposed)
The peptide is designed to antagonise α-melanocyte-stimulating hormone (α-MSH) at the MC1R receptor on melanocytes. α-MSH binding to MC1R is one of the main signals that drives melanocytes toward eumelanin production; blocking it is intended to reduce the instruction to pigment rather than chelating copper or jamming tyrosinase after the fact. It is a genuinely different and more upstream target than kojic acid, arbutin or decapeptide-12 — conceptually the same receptor family that afamelanotide activates, aimed in the opposite direction. The honest caveats are the familiar pair: receptor antagonism in a cell model is not a demonstrated skin outcome, and topical peptide penetration is uncertain.
Reading the evidence honestly
The clinical record is small and largely supplier-associated — manufacturer dossiers and short evaluations reporting reduced pigment appearance and more even tone. Independent human replication is limited, which is why the grade sits at D despite an interesting mechanism. As always, the endpoints concern the appearance of existing pigment, not a change in baseline skin colour — the line that separates an honest brightening claim from a whitening one.
