Identity
Tripeptide-29 is a synthetic cosmetic peptide used under the INCI name Tripeptide-29. Its design is biomimetic: it is built on the Gly-Pro-Hyp repeat, the very motif whose repetition forms the triple helix of collagen itself. That places it in a different conceptual box from the matrikine signal peptides — instead of telling fibroblasts to build matrix (Matrixyl, Pal-GHK), it is a collagen-like fragment.
Mechanism (as proposed)
The premise is that a collagen-mimetic peptide either integrates with or supports the existing matrix, or signals repair by resembling collagen fragments. It is an appealing idea — a small piece shaped like the thing it is meant to support. But mechanistically it is less characterised than the matrikines, and the two honest gaps are large: a collagen-like sequence applied topically has not been shown to raise collagen in human skin, and whether it penetrates intact skin at all is uncertain. The concept is doing more work here than the data.
Reading the evidence honestly
The human record is minimal and largely supplier-associated — even thinner than the signal peptides, which at least have Matrixyl's split-face RCT to anchor the class. Independent replication is essentially absent, which is why the grade sits at D across the board. What evidence exists concerns appearance, not collagen content, so the "collagen-boosting" framing is a structural claim the data does not support.
