Identity
Teprenone (INCI; also geranylgeranylacetone, GGA; trademarked Renovage by Sederma) is a small acyclic terpenoid ketone. Before cosmetics it was — and still is — a prescription anti-ulcer drug in Japan. As a cosmetic it is sold on a telomere / "cellular anti-ageing" story that its evidence does not carry.
The molecule was a stomach drug first
Under the name Selbex, geranylgeranylacetone has been prescribed in Japan since 1984 for gastritis and stomach ulcers, working by coaxing cells to make heat-shock proteins — cellular first-responders that protect the stomach lining. Solid, unglamorous, real medicine. That is the raw material that was later carried out of the gut and onto the face.
Enter Sederma, and a very big word
Around 2007 the French cosmetic-ingredient house Sederma dissolved teprenone in a light carrier oil, trademarked it Renovage, and built the story around the most seductive word in ageing science: telomeres — the protective caps on chromosome ends that shorten as cells divide. A topical that "protects the telomeres of skin cells" sounds like it strikes at the root of ageing. It is a wonderful story. Look at what sits underneath it.
The single study, and the hole in the middle of it
The evidence for Renovage is essentially one clinical study, produced by the company selling the ingredient — about 24 volunteers, average age ~58, 3% teprenone. The instrument readings are the honest part: firmness up roughly 35%, tone up, pore size and redness down, pigmented spots reduced over six months. If that were the whole claim it would be an ordinary, defensible cosmetic result (its C).
But the study was built to support a telomere claim, and here is the hole: it did not measure telomere length. Nothing in it shows a telomere getting longer, or staying longer, in human skin, and no independent laboratory has since shown a topical teprenone altering telomeres, telomerase, or cell lifespan in living skin. The word doing all the marketing work is the one thing the trial never checked — a textbook, unusually ambitious outcome substitution, firewalled at F.
What teprenone can honestly be graded on
- The look of firmness, tone and pores in mature skin — C. Plausible, but the whole case is one unblinded, manufacturer-run trial.
- Antioxidant / environmental-defence appearance — C. Real antioxidant / heat-shock biology; modest, lightly-evidenced on-skin payoff.
- The look of dark spots — D. A single lab finding plus one in-house "-56%" figure is not a proven brightening active.
- "Lengthens telomeres / renewal at the DNA level" — F. The headline claim and the least supported thing about the ingredient.
The test you can reuse
Did any study actually measure the mechanism in the claim — or only how skin looked? If a serum promises telomere protection and the trial behind it measured firmness and pore size in the maker's own lab, the mechanism is decoration.
