Short answer: no study has ever shown it. Niacinamide is one of the best-evidenced actives in skincare – but "boosts collagen" describes a mechanism its trials never measured. Graded D as a claim, not because the ingredient is weak, but because the word on the box never appears in the science it points to.
Here is the strange part. Go and read the papers that made niacinamide famous. The word "collagen" is not in them.
The story starts around the year 2000, in two laboratories on opposite sides of the world, inside the same company.
In Kobe, Japan, a Procter & Gamble scientist named Tomohiro Hakozaki was chasing a specific problem. Across much of the Asian market, customers wanted an even, lighter-looking complexion, and the industry had no gentle, well-tolerated ingredient that reliably delivered it. Hakozaki thought a humble B-vitamin might. Half a world away, at P&G's Miami Valley laboratories outside Cincinnati, a veteran researcher named Donald Bissett was running the human trials that would decide whether the hunch held up.
It held up. In 2002, Hakozaki and his colleagues published the study that changed the ingredient's life. Working with cultured cells and with eighteen Japanese women in a split-face trial – niacinamide on one cheek, plain moisturiser on the other – they showed something clean and specific. Niacinamide suppressed the transfer of pigment packets, called melanosomes, from the cells that make pigment to the cells that carry it to the surface. In the lab models the drop was between thirty-five and sixty-eight percent. On the face, the treated side grew visibly, measurably lighter.
That is a real discovery. It is why niacinamide is in half the serums on the shelf today. And notice what it is a discovery about: the appearance of pigmentation. Not collagen.
Bissett's follow-up work, published in 2004 and 2005, widened the picture. His twelve-week trials found niacinamide reduced yellowing, the look of wrinkles, red blotchiness, and hyperpigmented spots in ageing skin. Impressive range. And look at the title he chose for the 2005 paper – a sentence written by one of the people who knew the data best:
"Niacinamide: a B vitamin that improves aging facial skin appearance."
Appearance. He measured how skin looked, and he said so, precisely. He did not write "boosts collagen," because he had not measured collagen. Neither had Hakozaki. In the entire founding body of work – the studies every "collagen-boosting" niacinamide serum ultimately rests on – nobody looked at collagen at all.
So where did the word come from?
Somewhere between the Cincinnati lab bench and the shelf, "improves the appearance of ageing skin" quietly became "boosts collagen." It is easy to see the temptation. Skin that looks firmer reads as more collagen, because collagen is the word everyone associates with firm skin. The leap feels harmless. It is not.
Skin can look firmer and smoother for reasons that have nothing to do with collagen synthesis: better hydration, a calmer barrier, less surface light-scattering, reduced redness. An appearance trial cannot tell those apart from a genuine collagen change – it was never built to. So "improves the look of fine lines" is earned, tested, real. "Boosts collagen" is a mechanism bolted on top of an outcome that never checked for it.
We call this shape outcome substitution: a real measurement of one thing, sold as a different thing. It is the most common move in cosmetic marketing, and it is not lying – the study is real, the improvement is real. It is a swap. Once you can see it, you cannot unsee it.
The quiet irony is that the scientists themselves did not make the swap. Hakozaki and Bissett were disciplined about what their data did and did not show. The word "collagen" was added later, by people who never ran a trial.
This is not a takedown, and that matters – the honest ledger runs both ways.
If you reached for niacinamide because you wanted more even-looking skin, the evidence is genuinely, unusually strong for a cosmetic active. You were sold a good ingredient. You were just sold it with the wrong sentence.
The same substitution logic sinks another popular claim. More is not better. The comparison was actually run in humans – 2%, 5%, 10% – and 10% did not beat 5%. Every controlled result sits between 2 and 6%. The extra strength buys flushing and tingling, not proportionate benefit. "High-strength 10%+" grades D for the cleanest possible reason: somebody checked, and the higher number lost.
You do not need a chemistry degree to catch outcome substitution. You need one question, and it works on any product on any shelf:
What did the study measure, and is it the same thing the box promises?
If the box names a mechanism – collagen synthesis, barrier repair, plumping from within – and the evidence underneath is an appearance study, those are two different claims wearing the same label. The appearance result may be perfectly real. The mechanism is an addition.
Claim "boosts collagen": grade D. The human data on niacinamide is strong and real – but it measured appearance, not collagen, so it cannot support this specific promise. The ingredient deserves its reputation. The sentence on the box does not. Buy niacinamide for how skin looks and feels; not for a collagen mechanism that the two scientists who made it famous never once claimed.
